De novo thymus regeneration in the axolotl

Although regeneration abilities are generally restricted among vertebrates, the axolotls are exceptional in their capacity to regenerate complete limbs post-amputation as well as large parts of various organs including the brain, spinal cord or heart. We have recently found that, even more extraordinarily, the axolotl can regenerate a complete organ – the thymus – /de novo/. The thymus is the primary organ of the adaptive immune system playing a crucial role in T-cell development and specialization. We use a combination of imaging, tissue transplantation and single-cell RNA sequencing (scRNA-seq) approaches to address which molecular players are crucial for thymus regeneration, which cell type gives rise to the new structure, and whether the regenerated organ is completely functional. Ultimately, this work will contribute to our understanding of the cellular and molecular basis for de novo thymus regeneration and maintenance, of both basic and translational relevance.
